目的观察白藜芦醇对高脂饮食大鼠心肌过氧化物酶增殖物激活受体(PPARγ)及其下游信号分子激活蛋白-1(AP-1)表达影响。方法雄性SD大鼠32只,随机分为对照、高脂、吡格列酮、白藜芦醇4组;分别喂饲普通、高脂饲料,灌胃给予吡格列酮、白藜芦醇10 mg/(kg.d),连续6周;分别检测心肌PPARγmRNA、PPARγ、AP-1蛋白表达变化。结果高脂组PPARγmRNA表达(0.36±0.04)较对照组(0.48±0.04)明显降低(P<0.05);白藜芦醇组(0.54±0.11)较高脂组表达明显升高(P<0.05);高脂组PPARγ蛋白表达(0.40±0.02)较对照组(0.52±0.02)明显降低(P<0.05),而AP-1表达明显升高(P<0.05);白藜芦醇组PPARγ表达较高脂组升高,AP-1表达较高脂组降低(P<0.05)。结论白藜芦醇对高脂状态下大鼠心肌PPARγ/AP-1信号转导通路有激活作用。
Objective To observe the effects of resveratrol on the expression of myocardial peroxisome proliferator - activated receptor (PPARγ) and its downstream activator of metalloproteinase - 1 (AP - 1) in rats fed high fat diet. Methods Thirty-two male Sprague-Dawley rats were randomly divided into 4 groups: control group, high fat group, pioglitazone group and resveratrol group. The rats were fed normal and high-fat diet respectively, and received intragastric administration of pioglitazone and resveratrol 10 mg / ) For 6 weeks. The changes of PPARγmRNA, PPARγ, AP-1 protein expression were detected. Results Compared with the control group (0.48 ± 0.04), the expression of PPARγmRNA in the hyperlipidemic group (0.36 ± 0.04) was significantly lower than that in the control group (0.54 ± 0.11) (P <0.05) ; The expression of PPARγ protein in high-fat diet group (0.40 ± 0.02) was significantly lower than that in control group (0.52 ± 0.02) (P <0.05), while the expression of AP-1 was significantly increased (P <0.05) In the high fat diet group, the expression of AP-1 was lower than the high fat diet group (P <0.05). Conclusion Resveratrol can activate PPARγ / AP-1 signal transduction pathway in myocardium of rats under high fat condition.
