目的 观察不同病程的糖尿病大鼠肾组织内糖基化终产物受体(RAG)mRNA的表达水平的改变。方法 采用逆转录聚合酶链式反应(RT-PCR)检测病程12周内RAGEmRNA水平。结果 诱发糖尿病大鼠2周时肾脏皮、髓质内RAGEmRNA表达无明显变化,4周后其表达不断地显著增加(P<0.05)。诱发8周、12周的糖尿病大鼠,其糖化血红蛋白(GHb)水平明显升高(P<0.001)。结论 糖尿病状态下,肾组织内RAGE的高表达可能与糖尿病肾病的发病有关,且可能是高血糖诱发AGEs形成的结果。
Objective To observe the changes of RAG mRNA expression in the kidney of diabetic rats with different course of disease. Methods Reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the RAGE mRNA level within 12 weeks. Results The expression of RAGEmRNA in renal cortex and medulla had no significant changes at 2 weeks in diabetic rats, and the expression of RAGEmRNA in the medulla increased markedly after 4 weeks (P <0.05). The levels of HbA1c significantly increased in diabetic rats induced for 8 weeks and 12 weeks (P <0.001). Conclusion In diabetes, the high expression of RAGE in renal tissues may be related to the pathogenesis of diabetic nephropathy and may be the result of hyperglycemia-induced AGEs formation.
