本文报道用MClB(Monochlorobimane)能特异性标记细胞内谷胱甘肽(GSH),并与其它GSH测定方法(Tietze’s)有定量关系。对人体细胞株内GSH的测定得到下列结论:1.MClB需高于50~100倍才能充分标记。2.完全标记所需时间为45分钟到一小时。3.对人和小鼠细胞株测定,得出此法与Tiet-ze’s法在定性上一致性较好,但在定量上则较差。资料表明,测定人体肿瘤活组织GSH含量时,必需与Tietze’s法、MClB的高压液相法和MClB的流式细胞测定结合使用,以说明肿瘤细胞的非均一性。
This paper reports the specific labeling of intracellular glutathione (GSH) with MClB (Monochlorobimane) and has a quantitative relationship with other Tietze’s. Determination of GSH in human cell lines obtained the following conclusions: 1.MClB need to be higher than 50 to 100 times in order to fully labeled. 2. The time required for complete labeling is 45 minutes to one hour. The determination of human and mouse cell lines, concluded that this method and Tiet-ze’s method in qualitative consistency is better, but the quantitative is poor. The data indicate that determining the GSH content of human living tumor tissue must be used in combination with the Tietze’s method, the high-pressure liquid phase method of MClB, and the flow cytometry of MClB to account for the heterogeneity of the tumor cells.
