目的:探讨裸鼠结直肠癌循环肿瘤细胞的体内成瘤和转移情况。方法:将荧光素酶标记的人结直肠癌HCT116-luc细胞接种于裸鼠盲肠壁的黏膜与基层间,建立裸鼠模型。以上皮细胞黏附分子(epithelial cell adhesion molecule,EpCAM)免疫磁珠法筛选裸鼠结直肠癌外周血循环肿瘤细胞,将EpCAM~+和EpCAM~-细胞分别注射裸鼠尾静脉,观察裸鼠成瘤及肝肺转移情况。结果:成功建立了结直肠癌裸鼠原位移植瘤模型。EpCAM~+循环肿瘤细胞在裸鼠体内的成瘤率为90%,并出现60%肺转移、40%肝转移和35%肝肺同时转移;而EpCAM~-循环肿瘤细胞不能在裸鼠体内成瘤,也未出现转移。成瘤率和转移率在2组间的差异均有统计学意义(P值均<0.01)。结论:EpCAM~+循环肿瘤细胞在裸鼠体内的成瘤情况良好,且易出现转移。
Objective: To investigate the in vivo tumorigenesis and metastasis of colorectal cancer circulating tumor cells in nude mice. Methods: Luciferase-labeled human colorectal cancer HCT116-luc cells were seeded into the mucosa and basal layer of the cecal wall of nude mice to establish a nude mouse model. EpCAM ~ + and EpCAM ~ - cells were injected into the tail vein of nude mice respectively by the epithelial cell adhesion molecule (EpCAM) immunomagnetic beads method to detect the circulating tumor cells in peripheral blood of colorectal cancer in nude mice. Liver and lung metastases. Results: The in situ xenograft model of colorectal cancer was successfully established. The tumorigenic rate of EpCAM ~ + tumor cells in nude mice was 90%, and 60% of lung metastases, 40% of liver metastases and 35% of liver and lung metastases simultaneously. However, EpCAM ~ - circulating tumor cells could not form in nude mice Neoplasm, nor metastasis. The rates of tumor formation and metastasis in the two groups were statistically significant (P <0.01). Conclusion: The tumorigenesis of EpCAM ~ + circulating tumor cells in nude mice is good and prone to metastasis.
