目的 观察正常人成纤维细胞老化过程中β 半乳糖苷酶的变化规律。 方法 取正常人二倍体成纤维细胞进行传代培养 ,以不同代次的细胞为实验对象 ,通过细胞生长曲线、群体倍增时间和 β 半乳糖苷酶染色等一系列检测方法 ,对成纤维细胞老化过程进行了观察。结果 随着成纤维细胞的连续传代培养 ,SA β Gal表达呈现从弱 (在年轻细胞中占 4 % )到强 (在老化细胞中占 6 0 % )的趋势。结论 为建立成纤维细胞衰老模型提供了实验依据
Objective To observe the changes of β-galactosidase during normal human fibroblasts aging. Methods Normal human diploid fibroblasts were subcultured. Cells of different generations were used as experimental subjects. A series of detection methods such as cell growth curve, population doubling time and β-galactosidase staining were used to detect the fibroblasts aging The process was observed. Results With continuous subculture of fibroblasts, SA [beta] Gal expression tended to be weak (from 4% in young cells) to strong (60% in aged cells). Conclusion This study provides an experimental basis for establishing a fibroblast senescence model
