基于线粒体DNA COI基因序列分析了浙江和福建沿海的厚壳贻贝Mytilus coruscus群体遗传结构及遗传多样性现状。研究结果显示:在长度为564bp的COI基因片段上,厚壳贻贝的5个群体均呈现较高的单倍型多样度(0.867±0.129~1.000士0.177)和较低的核苷酸多样度(0.007±0.005~0.011±0.008);邻接关系树显示,5个群体的单倍型相互混杂,仅有1个小的分支存在;厚壳贻贝单倍型网络关系图呈星状结构,存在1个与系统树相对应的分支;不同群体间的F_(ST)值较小,且统计检验的结果均不显著,表明浙江、福建沿岸的厚壳贻贝群体问在COI基因上存在同质性;Tajima’s D和Fu’s F_s中性检验的结果(Tajima’s D=-1.828,P=0.017;Fs=-22.954,P=0.000)都显著偏离中性,提示厚壳贻贝经历了群体扩张。
Based on the mitochondrial DNA COI gene sequences, the genetic structure and genetic diversity of Mytilus coruscus populations in the coastal area of Zhejiang and Fujian were analyzed. The results showed that in the COI gene fragment with a length of 564bp, the five populations of M. mussels exhibited higher haplotype diversity (0.867 ± 0.129 ~ 1.000 ± 0.177) and lower nucleotide diversity (0.007 ± 0.005 ~ 0.011 ± 0.008). The contig relationship tree showed that the haplotypes of the five populations were mixed with each other and only one branch was present. The haplotype network of thick-shell mussel showed star-like structure and existed 1 branch corresponding to the phylogenetic tree. The F_ (ST) values of different groups are small, and the results of statistical test are not significant, indicating that there exists homogeneity in the COI gene of the mussel shellfish populations in Zhejiang and Fujian coasts (Tajima’s D = -1.828, P = 0.017; Fs = -22.954, P = 0.000) showed significant deviations from neutrality, suggesting that the mussel shell had undergone population expansion.
