目的:探讨槲皮素联合顺铂对HeLa细胞增殖及凋亡的影响。方法:不同浓度槲皮素及不同浓度槲皮素联合顺铂处理HeLa细胞24 h后,采用MTT检测细胞的增殖活性、流式细胞仪Annexin V-FITC/PI法检测细胞的凋亡情况、激光扫描共聚焦显微镜观察用PI荧光标记的细胞核的形态学变化。结果:槲皮素能抑制HeLa细胞增殖,且呈剂量依赖性;槲皮素联合顺铂与相同浓度顺铂单用处理HeLa细胞相比,前者对HeLa细胞的抑制作用更显著(P<0.01);不同浓度槲皮素与顺铂联合处理HeLa细胞与顺铂单用相比,前者显著提高了HeLa细胞的凋亡率(P<0.01)。结论:槲皮素可显著抑制HeLa细胞增殖并诱导其凋亡,槲皮素与顺铂联合应用具有一定的协同效应。
Objective: To investigate the effect of quercetin and cisplatin on the proliferation and apoptosis of HeLa cells. Methods: HeLa cells were treated with different concentrations of quercetin and different concentrations of quercetin and cisplatin for 24 h. MTT was used to detect the proliferation of HeLa cells. The apoptosis of HeLa cells was detected by flow cytometry Annexin V-FITC / PI. Scanning confocal microscopy was used to observe morphological changes of PI-fluorescently labeled nuclei. Results: Quercetin inhibited the proliferation of HeLa cells in a dose-dependent manner. Quercetin combined with cisplatin showed more significant inhibitory effect on HeLa cells than HeLa cells treated with cisplatin alone at the same concentration (P <0.01) HeLa cells treated with different concentrations of quercetin and cisplatin significantly increased the apoptosis rate of HeLa cells compared with cisplatin alone (P <0.01). Conclusion: Quercetin can significantly inhibit the proliferation and induce apoptosis of HeLa cells. Combination of quercetin and cisplatin has a synergistic effect.
