目的研究吉林省2009~2010年麻疹病毒流行株的核蛋白(Nucleoprotein,NP)基因亲缘关系及氨基酸变异。方法对吉林省2009~2010年麻疹病毒流行株,用逆转录-聚合酶链反应扩增NP基因片段后测序,分析其NP基因亲缘关系及其氨基酸变异。结果基因亲缘关系树显示,吉林省2001~2010年47株麻疹病毒均为H1基因型,2009~2010年92.1%(35/38)的麻疹病毒流行株处于一个相对独立的分支。由NP基因序列推导的氨基酸序列分析显示,该92.1%的麻疹病毒流行株均有共同的3个氨基酸位点呈现变异。结论吉林省2009~2010年92.1%的麻疹病毒流行株呈现氨基酸变异,提示要关注NP氨基酸变异趋势。
Objective To study the genetic relationship and amino acid variation of the nucleoprotein (NP) gene of measles virus from 2009 to 2010 in Jilin Province. Methods The NP gene fragment was amplified by reverse transcription - polymerase chain reaction (PCR) from 2009 to 2010 in Jilin Province. The NP gene was sequenced and its genetic relationship and amino acid variation were analyzed. Results The genetic relationship tree showed that 47 measles viruses were H1 genotype in Jilin Province from 2001 to 2010, and 92.1% (35/38) measles virus epidemic strains were in a relatively independent branch from 2009 to 2010. The amino acid sequence deduced from the NP gene sequence showed that the 92.1% of measles virus strains shared three common amino acid mutations. Conclusion 92.1% of the measles virus strains present in Jilin Province from 2009 to 2010 showed amino acid variation, suggesting that attention should be paid to the variation trend of NP amino acids.
