目的 探讨白细胞介素1-β转化酶(ICE)在局灶性脑缺血再灌注后的表达及作用。方法 线栓法复制大脑中动脉脑缺血再灌注模型,逆转录聚合酶链式反应(RT-PCR)技术检测大鼠局灶性脑缺血再灌注后ICE mRNA表达。结果 缺血3h及缺血3h再灌注,随缺血及缺血再灌注时间延长,缺血中心区与半影区ICE mRNA表达处于动态变化之中,再灌注24h、48h半影区表达持续高水平,而中心区表达下降。结论 局灶性脑缺血再灌注过程中ICE mRNA表达增强,促进神经细胞凋亡,ICE参与局灶性脑缺血再灌注神经细胞凋亡的调控。
Objective To investigate the expression of interleukin 1-β converting enzyme (ICE) after focal cerebral ischemia-reperfusion in rats. Methods The middle cerebral artery occlusion (MCAO) model of cerebral ischemia-reperfusion was established by thread occlusion method. The expression of ICE mRNA was detected by reverse transcription-polymerase chain reaction (RT-PCR) after focal cerebral ischemia-reperfusion in rats. The results showed that ICE mRNA expression in the ischemic center and penumbra was in a dynamic state at 3h after ischemia and 3h after ischemia reperfusion. With the prolongation of ischemia and reperfusion, Level, while the central area decreased expression. Conclusion The expression of ICE mRNA increases during focal cerebral ischemia-reperfusion, promotes the apoptosis of nerve cells and ICE participates in the regulation of neuronal apoptosis after focal cerebral ischemia-reperfusion.
