目的:研究南方红豆杉种子萌发的机制,即南方红豆杉种子形态后熟过程中的生物大分子的变化过程。方法:采用0.05%赤霉素(GA3)加低(4℃)-暖(23℃)-低(4℃)变温层积处理南方红豆杉种子,研究其生理生化变化。结果:采用激素加变温层积法处理种子后,蛋白质、氨基酸、可溶性糖都具有在低温层积时期累积较快,暖层积时期消耗较快的特点,三者的含量总体都呈上升趋势,分别比处理前增加115.56%、123.70%、50.71%,但脂肪含量下降40.00%。脱氢酶、过氧化物酶(POD)、多酚氧化酶(PPO)、蛋白酶活性分别比处理前增加111.05%、178.73%、141.84%、169.97%。结论:激素加变温层积法影响南方红豆杉种子生理生化变化,使种子内贮藏物质不断分解代谢、转化和利用,这也是破除其休眠的关键。
Objective: To study the mechanism of seed germination of Taxus chinensis var. Mairei, that is, the process of biomacromolecule during the ripening of Taxus chinensis var.amuratio. Methods: Taxus chinensis seeds were treated with 0.05% GA3, cold (4 ℃) -heat (23 ℃) -low (4 ℃), and their physiological and biochemical changes were studied. Results: The contents of protein, amino acid and soluble sugar accumulated faster during the cold stratification period and consumed faster during the warm stratification period when the seeds were treated with the hormone plus temperature stratification method. The contents of the three components all showed an upward trend, Respectively increased by 115.56%, 123.70% and 50.71% than before treatment, but the fat content decreased by 40.00%. The activities of dehydrogenase, peroxidase (POD), polyphenol oxidase (PPO) and protease increased by 111.05%, 178.73%, 141.84% and 169.97%, respectively. CONCLUSION: The hormones and temperature-changing stratification method affect the physiological and biochemical changes of Taxus chinensis var.mongolica seed, and the catabolism, transformation and utilization of the stored materials in the seed are continuously broken down, which is also the key to breaking the dormancy.
