目的探讨血管紧张素Ⅱ(AngiotensinⅡ,AngⅡ)对体外培养人肾小球系膜细胞(human glomerular mesangial cells,HMC)生长的影响及荔枝核皂苷的干预作用。方法体外培养HMC细胞,用四甲基偶氮唑蓝(MTT)法进行AngⅡ有效作用浓度筛选和荔枝核皂苷的干预研究,确定AngⅡ作用的最佳浓度及荔枝核皂苷的干预作用;ELISA法检测细胞分泌IL-6、IL-1β的含量,观察荔枝核皂苷对其影响。结果 MTT法检测结果显示,AngⅡ作用于HMC细胞后,吸光度值(OD值)明显增加(P<0.05),10-6mol/ml AngⅡ在48h作用最明显(P<0.01)。ELISA法检测结果显示,荔枝核皂苷可以抑制HMC细胞的增殖,降低IL-6和IL-1β的水平,与AngⅡ组比较有显著性差异(P<0.01)。结论 AngⅡ可诱导HMC细胞增殖,10-6mol/ml在48h作用最明显;荔枝核皂苷可以降低HMC细胞分泌IL-6和IL-1β的含量,抑制HMC细胞的增殖,减少细胞外基质,从而延缓与减轻肾小球硬化。
Objective To investigate the effect of Angiotensin Ⅱ (AngⅡ) on the growth of human glomerular mesangial cells (HMC) in vitro and the effect of litchi saponin. Methods HMC cells were cultured in vitro, and the concentration of AngⅡ and the intervention of litchi saponin were determined by MTT assay to determine the optimal concentration of AngⅡ and the intervention effect of litchi saponin. ELISA assay The levels of IL-6 and IL-1β secreted by cells were observed, and the effects of lychee nuclear saponin on them were observed. Results The results of MTT assay showed that the absorbance value (OD) of HMC cells increased significantly after AngⅡ treatment (P <0.05), and the effect of Ang Ⅱ at 10-6mol / ml was most obvious at 48h (P <0.01). The results of ELISA showed that lychee nuclear saponin could inhibit the proliferation of HMC cells and decrease the levels of IL-6 and IL-1β, which were significantly different from those of AngⅡ group (P <0.01). Conclusion AngⅡ can induce the proliferation of HMC cells, and the most obvious effect is at 10-6mol / ml at 48h. Lychee nuclear saponin can reduce the levels of IL-6 and IL-1β secreted by HMC, inhibit the proliferation of HMC and decrease the extracellular matrix, And relieve glomerulosclerosis.
