目的:探讨脑衍生的神经营养因子(BDNF)及其受体trk B在老年性大鼠耳蜗中的表达。方法:采用实时定量PCR的方法对正常老年大鼠(老年非耳聋组)与老年性耳聋大鼠(老年耳聋组)及非老年正常大鼠(对照组)耳蜗组织中的BDNF和trk BmRNA表达进行定量分析。结果:定量分析结果显示,老年耳聋组耳蜗神经元和毛细胞中BDNF的表达比对照组明显减少(P<0.01);老年非耳聋组BDNF的表达与对照组比较,差异无统计学意义(P>0.05)。表达于神经元的trkB在老年耳聋组、老年非耳聋组和对照组差异均无统计学意义(P>0.05)。结论:老年性大鼠耳蜗中BDNF的表达与老年性大鼠耳聋的发病机制可能密切相关。
Objective: To investigate the expression of brain-derived neurotrophic factor (BDNF) and its receptor trk B in the cochlea of aged rats. Methods: BDNF and trk B mRNA expression in cochleas of normal aged rats (aged non-deafness group) and senile deafness rats (aged deafness group) and non-elderly normal rats (control group) were detected by real-time PCR Quantitative analysis. Results: The results of quantitative analysis showed that the expression of BDNF in cochlear neurons and hair cells in aged deafness group was significantly lower than that in control group (P <0.01). The expression of BDNF in aged non-deafness group was not significantly different from that in control group > 0.05). The expression of trkB in neurons was not significantly different between senile deafness group, senile deafness group and control group (P> 0.05). Conclusion: The expression of BDNF in senile rat cochlea may be closely related to the pathogenesis of senile deafness.
