目的探讨肾络通药物血清对大鼠系膜细胞外基质(ECM)不同成分的抑制作用及对结缔组织生长因子(CTGF)的影响。方法以体外培养大鼠肾小球系膜细胞及血清药理学方法为基础,采用酶联免疫吸附试验检测细胞外基质中纤维连接蛋白(FN)、Ⅳ型胶原(ColⅡ)的含量;采用逆转录-多聚酶链反应(RT-PCR)检测结缔组织生长因子mRNA的表达。结果细胞外基质纤维连接蛋白、Ⅳ型胶原的含量与结缔组织生长因子mRNA的表达呈正相关关系,肾络通对其均有抑制作用。结论肾络通能够减少细胞外基质的积聚,抑制结缔组织生长因子的分泌与表达,此作用是其防治肾小球硬化的可能机制之一。
Objective To investigate the inhibitory effect of Shenluotong drug serum on different components of rat mesangial extracellular matrix (ECM) and its effect on connective tissue growth factor (CTGF). Methods Based on the in vitro culture of glomerular mesangial cells and serum pharmacology, the content of fibronectin (FN) and type IV collagen (Col II) in extracellular matrix was detected by enzyme-linked immunosorbent assay (ELISA); - Polymerase chain reaction (RT-PCR) detects the expression of connective tissue growth factor mRNA. Results There was a positive correlation between the content of extracellular matrix fibronectin and type IV collagen and the expression of connective tissue growth factor mRNA, and both of them had inhibitory effects on them. Conclusion Shenluotong can reduce the accumulation of extracellular matrix and inhibit the secretion and expression of connective tissue growth factor, which is one of the possible mechanisms of prevention and treatment of glomerulosclerosis.
