目的研究苦丁茶中异绿原酸类活性成分对实验性2型糖尿病小鼠的降血糖作用。方法采用HP50SS小孔吸附树脂制备苦丁茶异绿原酸活性部位;采用高分辨液相串联质谱联用仪(LC-QTOF-MS/MS)分析该部位化学成分组成;采用高脂饲料配合腹腔注射链脲佐菌素的方式构建实验性2型糖尿病小鼠模型;以25,50,100 mg·kg~(-1)·d~(-1)该部位成分连续灌胃给药4周,观察糖尿病小鼠血清空腹血糖、葡萄糖耐受量、总胆固醇、低密度脂蛋白、超氧化物歧化酶、丙二醛指标的改善情况;观察小鼠肝脏油红O染色后的病理切片。结果苦丁茶中异绿原酸部位能够显著改善实验性2型糖尿病小鼠血清血糖、血脂相关指标,同时对小鼠肝脏的脂肪聚集有明细的抑制作用。结论苦丁茶中异绿原酸部位能显著改善实验性2型糖尿病小鼠的糖脂代谢紊乱。
Objective To study the hypoglycemic effects of different chlorogenic acids in Kudingcha on experimental type 2 diabetic mice. Methods HP50SS small hole resin was used to prepare the active site of Ilex kudingcha. LC-QTOF-MS / MS was used to analyze the chemical composition of this site. The model of experimental type 2 diabetic mice was established by injection of streptozotocin (STZ), and the components of this site were continuously gavaged for 4 weeks at 25, 50 and 100 mg · kg -1 (-1) d -1. Diabetes Fasting blood glucose, glucose tolerance, total cholesterol, low density lipoprotein, superoxide dismutase and malondialdehyde in mice were observed. The pathological sections of liver were observed after oil red O staining. Results The results showed that the different chlorogenic acid sites in Kudingcha could significantly improve the blood glucose and serum lipids in experimental type 2 diabetic mice and inhibit the liver fat accumulation in detail. Conclusion The different chlorogenic acid sites in Kudingcha could significantly improve the dyslipidemia in experimental type 2 diabetic mice.
