目的对Yam1B细胞自发产生的一种可抑制人T、B细胞增殖的可溶性因子(Yam-SF)进行研究。方法3H-TdR掺入法进行淋巴细胞增殖抑制测定,ELISA法测定免疫球蛋白,间接免疫荧光法测定IL-2R,CTLL-2细胞3H-TdR掺入法测定IL-2活性。结果Yam-1B培养上清(Yam-SF)经pronase水解消化而失活,经56℃处理10分钟,或在pH10环境中处理6小时抑制活性丧失,在pH2.0环境中可保持其抑制活性。凝胶过滤试验表明,培养上清抑制活性分布在分子量43000~67000范围内,且TGF-β抗体不能阻断其抑制活性。结论Yam-SF可能是一种新的免疫调节因子,该因子可抑制人T细胞及B细胞的增殖,并抑制正常B细胞免疫球蛋白的产生,但对PHA剌激的人淋巴细胞IL-2的产生以及IL-2受体(IL-2R)的表达无抑制作用。
Objective To study a spontaneously soluble factor (Yam-SF) that inhibits the proliferation of human T and B cells in Yam1B cells. Methods 3H-TdR incorporation method was used to determine the inhibition of lymphocyte proliferation. Immunoglobulin was detected by ELISA. IL-2 activity was measured by 3H-TdR incorporation assay of IL-2R and CTLL-2 cells by indirect immunofluorescence. Results The Yam-SF culture supernatant (Yam-SF) was inactivated by pronase hydrolysis and digestion, and its inhibitory activity was lost after it was treated at 56 ℃ for 10 minutes or at pH10 for 6 hours, and its inhibitory activity was maintained at pH2.0 . Gel filtration tests showed that the inhibitory activity of the culture supernatant was distributed in the range of molecular weight between 43000 and 67000, and the TGF-β antibody could not block its inhibitory activity. Conclusion Yam-SF may be a new immunoregulatory factor that inhibits the proliferation of human T cells and B cells and inhibits the production of normal B cell immunoglobulins. However, it is not sensitive to PHA-stimulated human lymphocytes IL-2 Production and IL-2 receptor (IL-2R) expression.
