目的研究4周游泳训练对db/db小鼠脂肪组织Netrin-1、UNC5B基因表达的影响。方法 db/db小鼠和同窝野生小鼠各16只随机分为野生对照组(WC)、db/db对照组(DC)、野生运动组(WE)和db/db运动组(DE),每组8只。4周游泳训练干预结束取附睾脂肪组织,RT-PCR法检测基因表达,Western Blotting法检测蛋白表达。结果与WC组相比,DC组小鼠附睾脂肪F4/80、CD68、TNF-α、IL-6、Netrin-1、UNC5B mRNA表达显著升高(P﹤0.05),F4/80、Netrin-1蛋白相对含量显著升高(P﹤0.05);与WC相比,WE组小鼠各项参数变化无显著差异;与DC组相比,DE组小鼠F4/80、CD68、TNF-α、IL-6、Netrin-1、UNC5B mRNA表达显著下降(P﹤0.05),F4/80、Netrin-1蛋白相对含量显著下降(P﹤0.05);小鼠附睾脂肪Netrin-1和F4/80蛋白相对含量显著相关(r=0.798 1,P﹤0.01)。结论2型糖尿病小鼠脂肪组织巨噬细胞相关炎症水平提高,同Netrin-1、UNC5B基因表达提高有关,4周游泳训练可能通过抑制Netrin-1/UNC5B信号缓解2型糖尿病小鼠脂肪组织巨噬细胞相关炎症。
Objective To investigate the effect of 4 weeks swimming training on the expression of Netrin-1 and UNC5B genes in db / db mouse adipose tissue. Conclusions The level of macrophage-associated inflammation in adipose tissue of type 2 diabetic mice is increased, which is related to the increased expression of Netrin-1 and UNC5B genes.
