目的探究基于Th1/Th2细胞平衡前泌通片对大鼠慢性非细菌性前列腺炎的治疗作用机理。方法 60只SD雄性大鼠随机分为空白组、模型组、阳性对照组、前泌通片低中高剂量组,每组10只。除空白组,余各组建立慢性非细菌性前列腺炎模型。建模成功后,阳性对照组给予前列通瘀胶囊混悬液灌胃,前泌通片低中高剂量组分别给予不同剂量前泌通混悬液灌胃,空白组给予生理盐水灌胃,连续灌胃4周,末次给药后24h,股动脉放血处死大鼠,光镜下观察前列腺组织病理改变,免疫组化法检测IFN-γ和IL-4并计算IFN-γ/IL-4比值。结果与模型组比,前泌通片高中剂量组组织炎症明显减轻,前泌通片高剂量组IFN-γ和IL-4含量显著降低(P<0.01),前泌通片中低剂量组IL-4含量显著降低(P<0.01),前泌通片高中低剂量组IFN-γ/IL-4比值显著升高(P<0.01)。结论前泌通片可减轻慢性非细菌性前列腺炎大鼠组织炎症和下调IFN-γ、IL-4含量,上调IFN-γ/IL-4比值。
Objective To investigate the therapeutic mechanism of Th1 / Th2 balance preequilization on chronic nonbacterial prostatitis in rats. Methods Sixty male Sprague-Dawley rats were randomly divided into blank group, model group, positive control group and pre-secretory tablet low-middle-high dose group, with 10 rats in each group. In addition to the blank group, Yu each group to establish chronic non-bacterial prostatitis model. After the model was established, the positive control group was given gavage of Qianlietongyu capsule, and the low, medium and high dose of Qianliantong tablet group was given gavage with different doses of prophylactoid and gavage respectively. The blank group was given gavage with saline and continuous irrigation The rats in the femoral artery were sacrificed at 4 weeks after the last administration, and the pathological changes of the prostate were observed under light microscope. The ratios of IFN-γ and IL-4 were measured by immunohistochemistry. Results Compared with the model group, the inflammation in high dose group of Qianbi Tong Tablet was significantly reduced, while the levels of IFN-γ and IL-4 in high dose of Qianpi Tong Tablet were significantly decreased (P <0.01) (P <0.01). The ratio of IFN-γ / IL-4 in the high-dose and low-dose Xianghuetong tablets group was significantly increased (P <0.01). Conclusions The former Xi Tong Tablet can reduce tissue inflammation and down-regulate the contents of IFN-γ and IL-4 in chronic non-bacterial prostatitis rats, and up-regulate the ratio of IFN-γ / IL-4.
