为了准确测定棉油皂脚中β-谷甾醇与菜油甾醇的含量,将皂脚通过预处理之后,采用反相C18柱,流动相为95:5(V/V)的甲醇水混合液,流速0.8mL/min,检测波长为210nm,外标法定量。结果表明β-谷甾醇在40.16~240.96μg/mL,菜油甾醇在30.02~181.2μg/mL有良好的线性关系,平均回收率分别为97.91%、98.11%。方法结果准确可靠,重现性好,适合于皂脚及其他植物下脚料中甾醇含量测定。
In order to accurately determine the content of β-sitosterol and campesterol in the soaps of cottonseed oil, the soapstock was pretreated by reversed-phase C18 column with a methanol-water mixture with a mobile phase of 95: 5 (V / V) 0.8mL / min, detection wavelength of 210nm, external standard method. The results showed that there was a good linear relationship between β-sitosterol at 40.16 ~ 240.96μg / mL and campesterol at 30.02 ~ 181.2μg / mL. The average recoveries were 97.91% and 98.11%, respectively. The method is accurate, reliable and reproducible. It is suitable for the determination of sterol content in soaps of other soapstocks and other plants.
