小鼠胸腺上皮细胞株 MTEC_1细胞与考的松耐受胸腺细胞(CRT)共同培养,在无任何外源刺激剂条件下,MTEC_1细胞可维持 CRT 存活,促进 CRT 增殖,双荧光抗体染色 FACS 分析证明,MTEC_1细胞选择性地支持 CD_4~+CD_8~-细胞增殖。在 Con A 活化条件下,MTEC_1细胞仍以促进 CD_4~+CD_8~-细胞增殖为主.MTEC_1细胞培养上清液(MTEC_1—SN)不能直接促进 CRT 增殖,但可维持其存活;在 Con A 活化条件下,则可促进 CRT 增殖。表型分析发现,MTEC_1—SN 对 CD_4~+CD_8~-和 CD_4~-CD_8~+细胞的存活和增殖无明显选择作用。说明 MTEC_1细胞对 CD_4~+CD_8~-细胞的选择作用是两种细胞直接作用的结果.
MTEC_1 cells were co-cultured with Cortisone-tolerant thymus cells (CRT). MTEC_1 cells could maintain the survival of CRT and promote the proliferation of CRT in the absence of any external stimuli. FACS analysis of double fluorescent antibody , MTEC_1 cells selectively support the proliferation of CD_4 ~ + CD_8 ~ - cells. Under the condition of Con A activation, MTEC_1 cells still promote the proliferation of CD_4 ~ + CD_8 ~ - cells.MTEC_1 cell culture supernatant (MTEC_1-SN) can not directly promote the proliferation of CRT, but can maintain its survival; Under the conditions, it can promote CRT proliferation. Phenotypic analysis found that MTEC_1-SN had no obvious effect on the survival and proliferation of CD_4 ~ + CD_8 ~ - and CD_4 ~ -CD_8 ~ + cells. It showed that the selection of MTEC_1 cells on CD_4 ~ + CD_8 ~ - cells was the direct result of both cells.
