目的观察通脉益智胶囊对ApoE基因敲除小鼠(ApoE~(-/-)小鼠)无名动脉易损动脉粥样硬化(AS)斑块是否具有稳定作用。方法8周龄的ApoE~(-/-)小鼠给予高脂饲料22周后,无名动脉已经形成易损AS斑块,将小鼠随机分为3组:通脉益智胶囊组、辛伐他汀组和模型组,另设6只正常C57BL/6小鼠作为对照组,分别给予720mg/(kg·d)通脉益智胶囊、2.8mg/(kg·d)辛伐他汀及生理盐水,8周后用免疫组化、Movat Pentachrome以及天狼星红染色观察各组AS斑块的形态变化。结果与模型组相比较,通脉益智胶囊组可显著降低AS斑块的面积、增加斑块胶原组织的含量(P<0.01、P<0.05),辛伐他汀组可显著增强AS斑块纤维帽的连续性、增加纤维帽的厚度(P<0.05)。结论通脉益智胶囊可通过减小AS面积以及增加斑块内部胶原组织,辛伐他汀可通过稳定斑块表面的纤维帽,分别达到稳定AS斑块的作用。
Objective To observe whether Tongmai Yizhi Capsule can stabilize atherosclerotic (AS) plaques in the innominate artery of ApoE knockout mice (ApoE~(-/-) mice). Methods Eight weeks old ApoE~(-/-) mice were given high-fat diet for 22 weeks. The innominate artery had formed vulnerable AS plaques. The mice were randomly divided into three groups: Tongmai Yizhi capsule group, Xinfa In the statin group and the model group, 6 normal C57BL/6 mice were also set as the control group and were given 720 mg/(kg·d) Tongmai Yizhi Capsule, 2.8 mg/(kg·d) simvastatin and saline, respectively. After 8 weeks, the morphology of AS plaques in each group was observed by immunohistochemistry, Movat Pentachrome and Sirius red staining. Results Compared with the model group, Tongmai Yizhi capsule group can significantly reduce the area of AS plaques and increase the content of plaque collagen tissue (P<0.01, P<0.05). Simvastatin group can significantly increase AS plaque fiber. Cap continuity, increased fibrous cap thickness (P < 0.05). Conclusion Tongmai Yizhi capsule can reduce the area of AS and increase the internal collagen of the plaque. Simvastatin can stabilize the plaque of AS by stabilizing the fibrous cap on the surface of the plaque.
