12周游泳运动对高脂饮食大鼠脂肪组织UCP2 mRNA和TNF-α的影响

目的:探讨脂肪组织UCP2mRNA和TNF-α在运动防治高脂饮食危害中的作用。方法:SD雄性大鼠30只随机分为普通饮食对照组(C)、高脂饮食对照组(H)、高脂饮食运动组(HE)和普通饮食运动组(E)。H组和HE组大鼠采用高脂饲料喂养;HE组和E组同时进行12周无负重游泳运动(90min/天,5次/周)。采用RT-PCR方法测试大鼠脂肪组织中UCP2mRNA和TNF-αmRNA表达水平,采用ELISA法测定大鼠脂肪组织和血清中TNF-α的含量。结果:(1)12周实验结束后,H组体重显著高于HE组和C组,但脂体比与C组比较差异不显著。HE组脂体比显著低于H组和C组;(2)H组大鼠血清TNF-α含量显著高于HE组和C组;HE组脂肪组织TNF-α含量与H组和C组比较差异不显著,E组脂肪组织中TNF-α含量显著低于HE组和C组;(3)H组大鼠脂肪组织中UCP2mRNA和TNF-αmRNA表达显著高于HE组和C组;HE组脂肪组织中UCP2mRNA和TNF-αmRNA显著高于C组和E组;E组和C组之间差异不显著。结论:高脂饮食大鼠脂肪组织TNF-αmRNA和UCP2mRNA升高,运动可降低高脂饮食大鼠脂肪组织TNF-αmRNA和UCP2mRNA升高的程度,这种调节变化可能在运动减少高脂饮食导致的脂肪累积中发挥一定的作用。 Objective: To investigate the role of adipose tissue UCP2 mRNA and TNF-α in the prevention and treatment of high-fat diet hazards. Methods: Thirty SD male rats were randomly divided into normal diet control group (C), high fat diet control group (H), high fat diet exercise group (HE) and normal diet exercise group (E). Rats in H group and HE group were fed with high-fat diet. Rats in HE group and E group received no-load swimming exercise at the same time for 12 weeks (90min / day, 5times / week). The expression of UCP2 mRNA and TNF-αmRNA in rat adipose tissue was detected by RT-PCR. The content of TNF-α in adipose tissue and serum of rats was determined by ELISA. Results: (1) At 12 weeks after the experiment, the body weight of H group was significantly higher than that of HE group and C group, but the difference of lipid ratio was not significant compared with C group. The body fat ratio in HE group was significantly lower than that in H group and C group. (2) The content of TNF-α in serum in H group was significantly higher than that in HE group and C group. The content of TNF-α in adipose tissue in HE group was significantly lower than that in H group and C group (3) The expression of UCP2 mRNA and TNF-αmRNA in adipose tissue of H group was significantly higher than that of HE group and C group. In HE group, the fat content of adipose tissue was significantly lower than that of HE group UCP2 mRNA and TNF-αmRNA in the tissue were significantly higher than those in the C and E groups. There was no significant difference between the E and C groups. CONCLUSIONS: The increase of TNF-αmRNA and UCP2mRNA in adipose tissue of rats fed with high-fat diet can reduce the increase of TNF-αmRNA and UCP2mRNA in adipose tissue of rats fed with high-fat diet, which might be caused by the reduction of exercise Fat accumulation in play a role.
