目的探讨华蟾素对体外培养的子宫内膜癌Ishikawa细胞形态、增殖、凋亡、周期分布及侵袭能力的影响。方法体外培养Ishikawa细胞,不同浓度的华蟾素干预后,光镜观察细胞形态改变,采用MTT法检测细胞的增殖,流式细胞术分析细胞凋亡及其周期,用Transwell小室检测细胞体外侵袭能力。结果华蟾素作用后细胞由梭形、多边形变成圆形,浮起。华蟾素能有效地抑制Ishikawa细胞的增殖,降低侵袭能力,诱导细胞凋亡,并使细胞周期阻滞于G1期,减少S和G2期的细胞。华蟾素终浓度为3.0mg/ml抑制作用最明显。结论华蟾素可抑制Ishikawa细胞增殖,降低侵袭能力,诱导细胞凋亡并阻断细胞周期。
Objective To investigate the effect of cinobufacini on the morphology, proliferation, apoptosis, cycle distribution and invasion of Ishikawa cells cultured in vitro. Methods Ishikawa cells were cultured in vitro. After intervention with different concentration of cinobufacini, the cell morphological changes were observed under light microscope. The proliferation of cells was detected by MTT assay. The apoptosis and cycle of cells were analyzed by flow cytometry. . Results After cinobufagin treatment, the cells changed from fusiform and polygons to round and floating. Cinobufacini can effectively inhibit the proliferation of Ishikawa cells, reduce the invasive ability, induce apoptosis, and arrest the cell cycle in G1 phase and decrease the cells in S and G2 phase. Cinobufagin final concentration of 3.0mg / ml the most obvious inhibition. Conclusion Cinobufacini can inhibit Ishikawa cell proliferation, reduce invasiveness, induce apoptosis and block cell cycle.
