目的:建立瓜馥木中总生物碱含量测定的方法。方法:用分光光度法测定瓜馥木中总生物碱的含量。以瓜馥木碱甲为对照品,溴甲酚绿为酸性染料,缓冲溶液pH值=4.8条件下,用8 mL氯仿萃取3次,于407 nm波长处测定其吸光度。结果:瓜馥木碱甲在3.16-18.96μg/L浓度范围内线性关系良好,回归方程A=0.0701C+0.0041,r=0.9989,精密度良好,RSD%=1.34%(n=5);样品溶液显色3.0 h内基本稳定,吸光度的RSD%=1.07%;本法测定的重复性良好,RSD%=1.34%;平均加样回收率为100.87%,RSD%=2.25%。结论:该法简便、准确,可用于瓜馥木中总生物碱的含量测定。
Objective: To establish a method for determination of total alkaloids in melon. Methods: The content of total alkaloids in melon wood was determined by spectrophotometry. Bromocresol green was taken as acid reference substance with guanidine methyl as reference substance, and its absorbance was measured at 407 nm under the condition of pH = 4.8 and extracted with 8 mL chloroform for 3 times. Results: There was a good linear relationship between the concentration of melphadine and 3.16-18.96μg / L, the regression equation A = 0.0701C + 0.0041, r = 0.9989, with good precision and RSD% = 1.34% (n = 5) The color of the solution was basically stable within 3.0 h. The RSD% of absorbance was 1.07%. The repeatability was good with RSD% = 1.34%. The average recovery was 100.87% and RSD% was 2.25%. Conclusion: This method is simple, accurate and can be used for the determination of total alkaloids in melon.
