目的:研究清热消积方对SPC-A-1肺癌细胞及其诱导的HUVEC内皮细胞表达VEGF、bFGF的影响,并探讨其抗肿瘤血管生成的部分作用机制。方法:以不同浓度的清热消积方药液干预SPC-A-1肺癌细胞及其诱导的HUVEC内皮细胞后,采用双抗体夹心ELISA法分别检测两种细胞上清液中VEGF、bFGF的表达量。结果:经清热消积方药液干预后,该两种细胞的VEGF、bFGF表达量均出现不同程度的下降,且与药物浓度呈负相关,具有良好的浓度依赖性(P<0.05)。结论:清热消积方抑制SPC-A-1肺癌细胞及其诱导的血管内皮细胞表达VEGF、bFGF,可能是其抗肿瘤血管生成的作用机制之一。
Objective: To investigate the effect of “Qingre Xiaoji Decoction” on the expression of VEGF and bFGF in SPC-A-1 lung cancer cells and its induced HUVECs and to explore the mechanism of its anti-tumor angiogenesis. Methods: The SPC-A-1 lung cancer cells and their HUVEC-induced endothelial cells were treated with different concentrations of Qingre Xiaoji prescription liquid. The expression of VEGF and bFGF in the two cell supernatants was detected by double antibody sandwich ELISA . Results: After the intervention of Qingre Xiaoji Prescription, the expression of VEGF and bFGF of the two kinds of cells all decreased to some extent, and was negatively correlated with the drug concentration, with a good concentration dependence (P <0.05). Conclusion: Digesting Recipe can inhibit the expression of VEGF and bFGF in SPC-A-1 lung cancer cells and its induced vascular endothelial cells, which may be one of its anti-tumor angiogenesis mechanisms.
