目的:比较雪莲组织中蛋白质的提取方法。方法:采用酚法、TCA/丙酮法、尿素法提取雪莲组织中蛋白质,所得蛋白质进行一维SDS-PAGE和双向电泳检测,通过比较电泳图谱确定雪莲组织中蛋白质的最佳提取方法。结果:酚法提取的雪莲组织蛋白质杂质量最少、纯度最高,一维SDS-PAGE和双向电泳图谱显示其具有较多的蛋白质点,且蛋白质种类较多。结论:3种提取方法均有一定的蛋白质损失,建议采用酚法从雪莲组织中提取蛋白质,为雪莲蛋白质组学研究提供参考。
Objective: To compare the protein extraction methods of lotus tissue. Methods: The protein in lotus leaf tissue was extracted by phenol method, TCA / acetone method and urea method. The obtained protein was detected by one-dimensional SDS-PAGE and two-dimensional electrophoresis. The optimal extraction method of protein in lotus tissue was determined by comparing the electrophoresis patterns. Results: The phenolic compounds extracted by phenolic method had the least amount of impurities and the highest purity. One-dimensional SDS-PAGE and two-dimensional electrophoresis showed that it had more protein spots and more protein types. CONCLUSION: All three extraction methods have some protein loss. It is suggested that the phenol extraction from the lotus tissue protein, for the Saussurea proteomics study provide a reference.
