在教学上,用容易观察的蛙卵做脊椎动物发生的观察材料是合适的。但蛙胚制片方法繁琐,卵黄易碎,往往得不到满意的效果。本文对蛙胚切片方法进行了一些改进,在解决卵黄易碎问题时,应用了在植物制片中颇受欢迎的脱水剂——正丁醇,其优点有二:(1)能与水、酒精及石蜡互溶,故脱水之后可不经过二甲苯透明而直接进行石蜡浸透、包埋;(2)很少引起组织块收缩和变脆。采用苏木精——伊红整染,故可供制作大量教学用片,既省时间又省药品,现将本法简介于下。
In teaching, it is appropriate to make observation materials for the occurrence of vertebrates with easily observed frog eggs. However, frog embryo production methods cumbersome, yolk fragile, often without satisfactory results. In this paper, some improvements have been made to the frog embryo slicing method. In the process of solving the problem of fragile yolk, the popular dehydrating agent n-butanol used in plant production has been applied. The advantages are twofold: (1) Alcohol and paraffin miscible, it can be dehydrated without xylene after direct paraffin penetration, embedded; (2) rarely cause tissue shrinkage and brittle. The use of hematoxylin - eosin dyeing, it is available for the production of a large number of teaching films, both save time and medicine, now this law brief below.
