DCC基因位于染色体18q21.3,是结直肠肿瘤发生发展中具有重要意义的抑癌基因。应用Southern印迹技术和限制性片断长度多态性(RFLP)研究了33例结直肠癌病人癌组织及其正常粘膜组织基因组DNA,其中16例,占49%具有杂合性,并检出6例,占38%存在18q等位基因杂合性缺失。18q等位基因杂合性缺失与肝癌和淋巴结转移有明显相关性。提示分子遗传学研究在结直肠肿瘤临床上如估计肿瘤转移能力有一定意义。
The DCC gene is located on chromosome 18q21.3 and is an important tumor suppressor gene in the development of colorectal tumors. Southern blotting technique and restriction fragment length polymorphism (RFLP) were used to study the genomic DNA of 33 cases of colorectal cancer patients and their normal mucosal tissues, of which 16 cases were heterozygous for 49%, and 6 cases were detected. 38% of the 18q alleles were missing. Loss of heterozygosity for the 18q allele was significantly associated with liver cancer and lymph node metastasis. It is suggested that molecular genetics studies have a certain significance in the clinical evaluation of colorectal tumors, such as estimating tumor metastasis.
