应用聚合酶链反应(PCR)放大口腔癌组织中H-ras癌基因部分DNA顺序,再以特异突变的寡脱氧核苷酸(ODN)探针杂交方法检测口腔癌中H-ras癌基因第12位密码子的G→T突变。结果在21例口腔癌中有6例存在这种突变(28.5%),而10例正常门腔粘膜组织中无1例发生突变。提示:H-ras癌基因第12位密码子→T突变可能是口腔癌中癌基因激活的方式之一。
The DNA sequence of H-ras oncogene in oral cancer tissues was amplified by polymerase chain reaction (PCR), and the mutation of H-ras oncogene in oral cancer was detected by specific mutated oligodeoxynucleotide (ODN) probe hybridization method G → T mutation of the codon. RESULTS: Sixteen of 21 oral carcinomas had this mutation (28.5%), whereas none of 10 normal mucosal tissues had mutations. Hint: H-ras oncogene codon 12 → T mutation may be one of the ways of oncogene activation in oral cancer.
