为了筛选药用成分高的优质何首乌中药材,以组织培养获得的淡绿色疏松何首乌愈伤组织为材料,将其分割成体积一致的愈伤组织团接种到添加5.0mg/L 6-BA+1.0mg/L NAA的MS培养基中,用15~20cm高的30W紫外灯照射不同时间,再用HPLC法检测其愈伤组织中的二苯乙烯苷含量。结果表明,何首乌愈伤组织经紫外光照射50min/d的二苯乙烯苷含量最高,为0.010 8%。因此,适当的紫外光照射可提高何首乌愈伤组织中的二苯乙烯苷含量。
In order to screen high quality Polygonum multiflorum medicinal materials with high medicinal composition, the light green loose Polygonum multiflorum callus tissue obtained from tissue culture was divided into the same volume of callus groups and inoculated into the medium with 5.0 mg / L 6-BA + 1.0 mg / L NAA MS medium, irradiated with 15 ~ 20cm high 30W UV lamp for different time, and the content of stilbene glycoside in callus was detected by HPLC. The results showed that the content of stilbene glycosides in Polygonum multiflorum callus irradiated by ultraviolet light for 50min / d was 0.010 8%. Therefore, appropriate UV irradiation can increase the content of stilbene glycoside in Polygonum multiflorum callus.
