目的:探讨盐酸小檗碱对抑制血管平滑肌细胞增殖的干预机制。方法:分离培养大鼠血管平滑肌细胞(vascular smooth muflcle cells,VSMC),并分为实验组,对照组及空白组;MTT法选择盐酸小檗碱干预浓度;MTT法和[~3H]-胸腺嘧啶核苷渗入法检测VsMc增殖率;westem blotting方法检测ERK1/2、Akt的磷酸化蛋白的变化。结果:VSMC实验组细胞增殖率与对照组相比明显下调,P<0.05;小檗碱干预后,VSMc细胞ERK1/2、Akt的磷酸化蛋白的表达明显下调,与对照组相比,P<0.05。结论:盐酸小檗碱对血管平滑肌细胞的增殖有一定的抑制作用,可能与其抑制ERK1/2、Akt等的表达有关。
Objective: To investigate the mechanism of berberine on the inhibition of vascular smooth muscle cell proliferation. Methods: Vascular smooth muscle cells (VSMCs) were isolated and cultured. The cells were divided into experimental group, control group and blank group. Berberine hydrochloride was selected by MTT method. MTT assay and [~ 3H] The proliferation rate of VsMc was detected by nucleoside infiltration and the phosphorylation of ERK1 / 2 and Akt was detected by westem blotting. Results: The proliferation rate of VSMC group was significantly lower than that of the control group (P <0.05). The expression of phosphorylated protein of ERK1 / 2 and Akt in VSMc group was significantly decreased after berberine treatment. Compared with the control group, P < 0.05. Conclusion: Berberine can inhibit the proliferation of vascular smooth muscle cells, which may be related to the inhibition of ERK1 / 2 and Akt expression.
