鱼腥草素钠对铜绿假单胞菌致肺气虚证大鼠β-防御素的影响

目的探究鱼腥草素钠(Sodium Houttuyfonate:SH)对呼吸道被膜菌铜绿假单胞菌(Pseudomonas aeruginosa:PA)致肺气虚证大鼠肺部β-防御素的影响。方法采用呼吸道被膜菌铜绿假单胞菌滴鼻法致大鼠呼吸道慢性感染,辅以寒冷疲劳刺激,诱发大鼠肺气虚证模型,用鱼腥草素钠干预,设不给药组为模型组和未感染大鼠作为正常对照组,8周后观察证候表现,镜下观察肺部组织病理改变,酶联免疫吸附法测定支气管灌洗液中β-防御素含量。结果模型组符合肺气虚证候表现。鱼腥草素钠组、阿奇霉素(azithromycin:AZM)阳性对照组及正常对照组均无明显的肺气虚症候表现。病理学检查显示,模型组的大鼠支气管粘膜有大量的脱落、炎症细胞浸润和肺气肿的发生,而药物治疗组及正常对照组无炎症反应,肺部支气管粘膜完好;模型组肺部灌洗液β-防御素含量明显增高,药物治疗组β-防御素增高不明显,两组相比差异显著(P<0.01),药物治疗组和正常对照组相比无显著差异(P>0.05)。结论β-防御素增高可以加剧肺部损伤,在铜绿假单胞菌感染大鼠过程中,鱼腥草素钠使大鼠气道β-防御素维持在正常水平,是抑制气道病变以及炎症反应从而干预肺气虚证的发生的重要因素之一。 Objective To investigate the effect of Sodium Houttuyfonate (SH) on β-defensin in the lung of rats with pulmonary qi deficiency syndrome induced by Pseudomonas aeruginosa (PA). Methods Chronic aerobic respiratory tract infection was induced by respiratory membrane-coated bacteria Pseudomonas aeruginosa nasal drops, supplemented by cold fatigue stimulation to induce lung qi deficiency syndrome in rats. Houttuynia cordata intervention was used to establish the model group And uninfected rats as normal control group. The symptoms were observed after 8 weeks. Pathological changes of lung tissue were observed under microscope. The content of β-defensin in bronchoalveolar lavage fluid was determined by enzyme-linked immunosorbent assay. Results The model group was consistent with the performance of lung deficiency syndrome. Sodium houttuyfonate group, azithromycin (azithromycin: AZM) positive control group and normal control group showed no symptoms of lung deficiency syndrome. Pathological examination showed that bronchial mucosa of the model group had a large number of shedding, infiltration of inflammatory cells and emphysema, while no inflammatory reaction and pulmonary bronchial mucosa were found in the drug-treated group and the normal control group. The content of β-defensin in the lotion was significantly higher than that in the drug-treated group (P <0.05), but there was no significant difference between the two groups (P <0.01) . Conclusion The increase of β-defensin can aggravate lung injury. During the process of Pseudomonas aeruginosa infection in rats, sodium houttuyfonate maintains the airway β-defensin at normal level in rats and is an inhibitory effect on airway pathology and inflammation Response and thus interfere with the occurrence of pulmonary qi deficiency one of the important factors.
