目的:探讨涤痰汤对帕金森病大鼠多巴胺神经细胞凋亡及相关基因表达的影响。方法:应用6-羟基多巴胺立体定向注射制作PD大鼠模型。将成功的PD模型随机分为模型组与化痰组,另外纳入正常对照组和假手术组,分别给予相应处理,共14天。用TUNEL测定各组大鼠的细胞凋亡数,免疫组化染色法测定Bc l-2基因和Bax基因的表达。结果:与模型组比较,化痰组细胞凋亡数明显减少,差异有显著意义(P<0.05);同时Bc l-2的表达明显增多,Bax的表达明显降低,差异均有显著意义(P<0.01)。结论涤痰汤通过增强Bc l-2和降低Bax的表达,减少PD大鼠的神经细胞凋亡而发挥保护神经元的作用。
Objective: To investigate the effect of Ditan Decoction on the apoptosis of dopamine neurons and the expression of related genes in Parkinson’s disease rats. Methods: PD rat model was made by stereotactic injection of 6-hydroxydopamine. The successful PD model was randomly divided into model group and phlegm group, another group was included in the normal control group and sham operation group, respectively, for the corresponding treatment, a total of 14 days. The number of apoptotic cells in each group was measured by TUNEL, and the expressions of Bcl-2 and Bax were detected by immunohistochemical staining. Results: Compared with the model group, the number of apoptotic cells in the phlegm group was significantly decreased (P <0.05), while the expression of Bcl-2 and Bax were significantly decreased (P <0.01). Conclusion Ditan Decoction can protect neurons by increasing the expression of Bcl-2, decreasing the expression of Bax and decreasing the apoptosis of neurons in PD rats.
