目的:建立HPLC-Flu法检测血浆中低浓度的美托洛尔。方法:采用YWG C_(18)色谱柱,流动相为甲醇-醋酸和醋酸钠缓冲液(45:55,含0.25%庚烷磺酸钠),以普萘洛尔为内标,检测器波长:EM 302 nm,EX 284 nm。结果:线性范围为2.5~80 ng·ml~(-1),最低检测浓度为2.5 ng·ml~(-1)。日内RSD均小于8%,但日间RSD较大。能检测口服6.25 mg,12.5 mg美托洛尔的心表患者的血药浓度。结论:该方法灵敏度高,方法简便,适用于临床药物监测。
Objective: To establish HPLC-Flu method to detect low concentration of metoprolol in plasma. Methods: YWG C 18 column was used. The mobile phase consisted of methanol - acetic acid and sodium acetate buffer (45:55, containing 0.25% sodium heptanesulfonate) and propranolol as internal standard. The detector wavelength was EM 302 nm, EX 284 nm. Results: The linear range was 2.5 ~ 80 ng · ml ~ (-1) and the lowest concentration was 2.5 ng · ml ~ (-1). Days RSD were less than 8%, but larger daytime RSD. Can detect oral 6.25 mg, 12.5 mg of metoprolol in patients with cardiac levels of plasma concentrations. Conclusion: This method has high sensitivity, simple method and is suitable for clinical drug monitoring.
