[目的 ]研究脾切除对猪血清致大鼠肝纤维化的影响。 [方法 ]雄性Wistar大鼠 37只 ,随机分为正常组、模型组、切脾组和假手术组。采用猪血清腹腔注射复制大鼠肝纤维化模型 ,观察大鼠肝脏病理与胶原染色 ,检测大鼠肝功能和肝脏羟脯氨酸 (Hyp)水平。[结果 ]猪血清腹腔注射 12周后 ,模型组、切脾组和假手术组大鼠肝脏有纤细至较粗的完整的纤维间隔形成。与正常组大鼠相比 ,模型组、切脾组和假手术组大鼠血清丙氨酸氨基转移酶(ALT)活性明显升高 [(46 .79± 4 .76 )、(5 2 .5 0± 4 .4 9)、(5 1.90± 3.4 7)U/L ,P <0 .0 5 ],白蛋白明显下降 [(2 8.0 3±1.4 1)、(2 7.86± 1.6 6 )、(2 8.82± 1.37) g/L ,P <0 .0 1,<0 .0 5 ],Hyp明显升高 [(30 3.77± 32 .5 9)、(332 .6 2±5 4 .0 6 )、(30 0 .17± 4 8.0 5 ) μg/ g湿肝重 ,P <0 .0 1],而总蛋白和总胆红素无明显变化。与模型组相比 ,切脾组ALT活性明显高于模型组 (P <0 .0 5 ) ,Hyp与模型组相比有增高的趋势。[结论 ]切除脾脏不能阻止猪血清致大鼠肝纤维化模型的形成。
[Objective] To study the effect of splenectomy on liver fibrosis induced by pig serum. [Methods] Thirty-seven male Wistar rats were randomly divided into normal group, model group, splenectomy group and sham operation group. Rat model of hepatic fibrosis was induced by intraperitoneal injection of porcine serum. Pathological and collagen staining of rat liver was observed. The liver function and hepatic hydroxyproline (Hyp) levels in rats were detected. [Results] After 12 weeks of intraperitoneal injection of porcine serum, the livers of rats in the model group, the splenectomy group and the sham operation group were formed with the slender and thick intact fiber septa. Serum alanine aminotransferase (ALT) activity in model group, splenectomy group and sham operation group was significantly higher than that in normal group [(46.79 ± 4.76), (52.5 Albumin significantly decreased (2 8.03 ± 1.4 1), (2 7.86 ± 1.6 6), (2 ± 4.14), (5 1.90 ± 3.4 7) U / L, 2 8.82 ± 1.37) g / L, P <0.01, <0.05, Hyp significantly increased [(30.377 ± 32.59), (332.6 ± 54.04) , (30 0 .17 ± 4 8.05) μg / g wet weight of liver, P <0.01], while the total protein and total bilirubin had no significant changes. Compared with the model group, ALT activity was significantly higher in the splenectomy group than in the model group (P <0.05), and Hyp had a tendency to increase compared with the model group. [Conclusion] Excision of the spleen can not prevent the formation of rat hepatic fibrosis model induced by pig serum.
