目的探讨抗坏血酸(AA)联合维生素D_3(VD_3)对豚鼠体内1,25-(OH)_2D_3水平和溃疡性结肠炎(UC)的影响。方法随机将32只豚鼠按体重分为4组,每组8只,对照组(120 mg AA+2.8μg VD_3/kg)、模型组(120 mg AA+2.8μg VD_3/kg)、高、低剂量AA组(240 mg AA+2.8μg VD_3/kg、8 mg AA+2.8μg VD_3/kg);每组豚鼠均进食不含AA和VD的特殊饲料,7W后,对照组豚鼠继续饮用蒸馏水,其他3组豚鼠自由饮用3%葡聚糖硫酸钠(DSS)溶液,3 d后处死豚鼠,检测相关指标。结果与对照组比较,模型组豚鼠结肠DAI评分、大体形态评分和组织病理学评分升高,血清25-(OH)D_3、1,25-(OH)_2D_3水平降低,肝脏CYP2R1和肾脏CYP27B1表达下降,血清IL-6和TNF-α水平[分别为(80.21±6.22)、(174.08±11.04)ng/m L]升高;与模型组比较,低剂量AA组结肠DAI评分和大体形态评分升高,血清25-(OH)D_3水平降低,血清IL-6和TNF-α水平[分别为(93.23±4.76)、(208.46±14.15)ng/m L]升高;与低剂量AA组比较,高剂量AA组结肠DAI评分和大体形态评分下降,血清25-(OH)D_3、1,25-(OH)_2D_3水平升高,肝脏CYP2R1和肾脏CYP27B1表达升高,血清IL-6和TNF-α水平[分别为(75.20±3.85)、(167.83±11.85)ng/m L]降低,差异均有统计学意义(P<0.05)。结论 AA在适宜范围内能够促进VD_3更充分地羟化为活性形式1,25-(OH)_2D_3,AA联合VD_3对DSS诱导的豚鼠UC具有一定干预作用。
Objective To investigate the effects of ascorbic acid (AA) combined with vitamin D_3 (VD_3) on 1,25- (OH) _2D_3 and ulcerative colitis (UC) in guinea pigs. Methods Thirty-two guinea pigs were randomly divided into 4 groups according to body weight: 8 rats in each group, control group (120 mg AA + 2.8 μg VD_3 / kg), model group (120 mg AA + 2.8 μg VD_3 / kg) AA group (240 mg AA +2.8 μg VD_3 / kg, 8 mg AA +2.8 μg VD_3 / kg). Results Compared with the control group, the expression of DAI score, gross morphology score and histopathological score of guinea pig in model group were decreased, the level of serum 25- (OH) D 3, 1, 25- (OH) 2D 3 was decreased and the expression of liver CYP2R1 and kidney CYP27B1 was decreased , Serum IL-6 and TNF-α levels [(80.21 ± 6.22) and (174.08 ± 11.04) ng / m L], respectively. Compared with the model group, the DAI score and general morphology score , The level of serum 25- (OH) D_3 decreased and the level of serum IL-6 and TNF-α increased (93.23 ± 4.76, (208.46 ± 14.15) ng / m L] The DAI score and general morphology score decreased in the dose AA group. The levels of serum 25- (OH) D_3, 1, 25- (OH) _2D_3 were increased, the expressions of CYP2R1 and CYP27B1 in liver were increased, and the levels of IL-6 and TNF- [(75.20 ± 3.85) and (167.83 ± 11.85) ng / m L, respectively], the differences were statistically significant (P <0.05). Conclusions AA could promote the more complete hydroxylation of VD_3 to the active form of 1,25- (OH) _2D_3 in the proper range. AA combined with VD_3 had certain intervention on DSS-induced UC in guinea pigs.
