目的检测Krüppel-like factor 4(KLF4)在小鼠舌黏膜癌变模型中的表达,探讨KLF4与口腔黏膜癌变的关系。方法选用C57BL/6J小鼠75只,其中阴性对照组15只,饮用纯净水;实验组小鼠60只,饮用4-硝基喹啉-1-氧化物(4NQO)水溶液(50μg/ml)。实验组分别在第8、12、16、24周末各处死15只小鼠,取舌组织,观察并记录标本的大体损害,将其固定在10%中性福尔马林溶液中,用于HE染色及KLF4免疫组织化学染色。结果在4NQO的诱导下,成功构建了小鼠舌黏膜癌变模型。HE染色结果显示,随着4NQO处理时间的延长,小鼠舌黏膜病变逐步加重,呈现出从单纯增生、异常增生到癌的变化。免疫组化结果显示,KLF4在口腔癌变过程中呈现动态变化,与正常口腔黏膜上皮相比,KLF4在单纯增生、异常增生上皮中的表达没有明显变化,但在鳞癌中,KLF4的表达量明显下降,具有显著性差异(P<0.01)。结论 KLF4可能在口腔黏膜上皮癌变过程中发挥抑癌基因的作用。
Objective To detect the expression of Krüppel-like factor 4 (KLF4) in mouse lingual mucosa carcinogenesis model and to explore the relationship between KLF4 and carcinogenesis of oral mucosa. Methods 75 C57BL / 6J mice were used, of which 15 were negative control group, drinking pure water. 60 mice in experimental group were given 4-nitroquinoline-1-oxide (4NQO) aqueous solution (50μg / ml). In the experimental group, 15 mice were sacrificed at the end of the 8th, 12th, 16th and 24th week respectively. The tongue tissue was taken and the gross damage of the specimen was observed. The specimens were fixed in 10% neutral formalin solution and used for HE Staining and KLF4 immunohistochemical staining. Results Under the induction of 4NQO, a mouse model of tongue carcinogenesis was constructed successfully. The results of HE staining showed that with the prolongation of 4NQO treatment time, the pathological changes of tongue mucosa in mice gradually increased, showing a change from simple hyperplasia and abnormal proliferation to cancer. The results of immunohistochemistry showed that KLF4 showed dynamic changes during oral carcinogenesis. Compared with normal oral mucosa epithelium, KLF4 expression did not change significantly in simple hyperplasia and dysplastic epithelium, but in squamous cell carcinoma, the expression of KLF4 was significantly Decreased, with significant difference (P <0.01). Conclusion KLF4 may play a role in tumor suppressor gene in carcinogenesis of oral mucosa.
