鱼腥草素钠预防生物被膜引起的早期肺气虚证的研究

目的研究鱼腥草素钠早期抗大鼠呼吸道生物被膜菌感染预防肺气虚证发生的作用。方法健康SD大鼠随机分为正常对照组、铜绿假单胞菌(Pseudomonas aeruginosa,Pa)标准株(ATCC27853)和铜绿假单胞菌临床株(AH16)分别包括模型组、鱼腥草素钠(SH)治疗组和阿奇霉素(AZM)阳性对照组。将呼吸道生物被膜菌铜绿假单胞菌标准株(Pa)和临床株(AH16)以滴鼻法感染大鼠,并辅以寒冷、疲劳刺激引起大鼠呼吸道慢性感染,诱导大鼠肺气虚证的发生。治疗组与阳性对照组于感染成功后分别用鱼腥草素钠及阿奇霉素治疗。观察各组证候表现、血气变化,酶联免疫吸附法测定肺泡灌洗液中白细胞介素-8(IL-8)的含量变化,HE染色镜下观察肺支气管组织器官病理改变。结果模型组的大鼠证状符合肺气虚证的证候表现;药物治疗组肺气虚证状不明显;血气分析表明,模型组大鼠血液中动脉氧分压(Partial pressure of oxygen,PO2)均显著下降,与正常对照组比较,P<0.05,或P<0.01,而二氧化碳分压(Partial pressure of carbon dioxide,PCO2)均显著升高与正常对照组比较(P<0.05);治疗组大鼠血液中PO2和PCO2变化不明显,与正常对照组比较P>0.05;模型组大鼠肺部灌洗液中的IL-8的含量均明显升高,与正常对照组比较,P<0.05,或P<0.01;病理学检查模型组肺及支气管炎症明显,肺部特征符合慢性支气管炎、肺气肿病理变化,鱼腥草素钠治疗组和阿奇霉素阳性大鼠肺部无病理变化出现。结论鱼腥草素钠早期干预铜绿假单胞菌生物被膜呼吸道感染的大鼠能预防肺气虚证的发生。 Objective To study the effect of sodium houttuyfonate early on preventing the infection of respiratory biofilm in rats against pulmonary qi deficiency syndrome. Methods Healthy SD rats were randomly divided into normal control group, Pseudomonas aeruginosa (Pa) standard strain (ATCC27853) and Pseudomonas aeruginosa clinical strain (AH16), including model group, sodium houttuyfonate SH) treatment group and azithromycin (AZM) positive control group. The respiratory biofilm strains Pseudomonas aeruginosa (Pa) and clinical strains (AH16) were infected intranasally in rats, supplemented by cold, fatigue caused by chronic respiratory tract infection in rats induced lung deficiency syndrome occur. The treatment group and the positive control group were treated with sodium houttuyfonate and azithromycin after successful infection. The changes of the expression of interleukin-8 (IL-8) in bronchoalveolar lavage fluid and the changes of bronchial organ histopathology were observed by HE staining. Results The syndromes of the rats in the model group were consistent with the syndromes of the lung-Qi deficiency syndrome. The symptoms of the lung-Qi deficiency were not obvious in the drug-treated group. The blood gas analysis showed that the partial pressure of oxygen (PO2) (P <0.05 or P <0.01), while the partial pressure of carbon dioxide (PCO2) increased significantly compared with that of the normal control group (P <0.05). The treatment group rats The levels of PO2 and PCO2 in the blood were not significantly different from those in the normal control group (P> 0.05). The contents of IL-8 in the lung lavage fluid in the model group were significantly higher than those in the normal control group (P <0.05) P <0.01; pathological examination model group lung and bronchial inflammation is obvious, the lung features consistent with chronic bronchitis, emphysema pathological changes, the sodium houttuyfonate treatment group and azithromycin-positive rat lung pathological changes appear. Conclusion Sodium houttuyfonate early intervention of Pseudomonas aeruginosa biofilm respiratory tract infection in rats can prevent the occurrence of pulmonary qi deficiency syndrome.
