研究了用99mTc直接标记抗体单价片段Fab’的方法与条件.IgG型多抗经无花果蛋白酶消化后得到F(ab')2双价片段,再经2-巯基乙胺还原为Fab'单价片段,与99mTc及亚锡葡萄糖酸钠药盒溶液混合保温,即可得到99mTc-Fab’标记物,标记率为85%-95%。这标记物在小鼠血中的清除速率明显高于完整抗体标记物99mTc-IgG.
The method and conditions for directly labeling the antibody monovalent fragment Fab ’with 99mTc were studied. IgG polyclonal antibody is digested by protease to obtain F (ab ’) 2 bivalent fragment, and then reduced to Fab’ monovalent fragment by 2-mercaptoethylamine, mixed with 99mTc and sodium stannous gluconate kit for incubation, then The 99mTc-Fab ’tag was obtained with a labeling rate of 85% -95%. The rate of clearance of this marker in mouse blood was significantly higher than that of the intact antibody marker 99mTc-IgG.
