目的研究紫草素对脂多糖诱导的新生大鼠急性肺损伤(ALI)的保护作用。方法 7d龄SD大鼠40只随机均分为四组,采用脂多糖5mg/kg腹腔注射建立ALI大鼠模型。A、B和C三组分别于建模前1h用紫草素50、25和12.5mg/kg腹腔注射,D组为模型对照。另取10只7d龄SD大鼠作为空白对照(E)组。6h后,用ELISA测定各组大鼠血清TNF-α和IL-10的表达,并获取肺组织,测定肺组织湿干重(W/D)比率,观察病理变化。结果与D组比较,紫草素预处理组大鼠ALI呈剂量依赖性减轻,肺组织W/D比率改善(P<0.05),抑制TNF-α表达,上调IL-10表达(P<0.05或P<0.01)。结论紫草素预处理对模型大鼠ALI具有保护作用,可能与抑制TNF-α表达和增加IL-10表达有关。
Objective To study the protective effect of shikonin on lipopolysaccharide-induced acute lung injury (ALI) in neonatal rats. Methods Forty SD SD rats were randomly divided into four groups. ALI rat model was established by intraperitoneal injection of lipopolysaccharide 5mg / kg. Groups A, B and C were intraperitoneally injected with shikonin at doses of 50, 25 and 12.5 mg / kg respectively 1 h before modeling, and group D was model control. Another 10 7-day-old SD rats as a blank control (E) group. Six hours later, the serum levels of TNF-α and IL-10 were measured by ELISA. The lung tissues were obtained and the ratio of wet to dry weight (W / D) was measured to observe the pathological changes. Results Compared with group D, the ALI in shikonin preconditioning group was relieved in a dose-dependent manner and the W / D ratio in lung tissue was improved (P <0.05), and inhibited the expression of TNF-α and up-regulated the expression of IL-10 P <0.01). Conclusion Shikonin preconditioning has a protective effect on ALI in model rats, which may be related to the inhibition of TNF-α expression and the increase of IL-10 expression.
