目的研究染色质解旋酶ALC1与转录因子EZH2在肝细胞癌(HCC)内的定位表达及相关性。方法本研究采用RT-PCR方法、细胞免疫荧光双标记及免疫组织化学等方法,从mRNA水平及蛋白质水平研究ALC1与EZH2在肝癌细胞及肝癌组织中的共表达,及相关性。结果肝癌细胞内可同时检测到ALC1与EZH2 mRNA表达,免疫荧光显示阳性表达产物均定位于胞核,肝癌HepG2细胞中ALC1与EZH2阳性表达率分别为78.0%和89.2%,肝癌Huh7细胞中ALC1与EZH2阳性表达率分别为53.6%和74.6%,两者在HepG2细胞中的共表达率为71.2%,Huh7细胞中两者共表达率为47.3%;肝癌组织芯片免疫组织化学检测结果显示肝癌组织中ALC1阳性表达与EZH2的阳性表达正相关(P<0.05),其共表达率为48.1%。结论各肝癌细胞系ALC1与EZH2的阳性表达率及共表达率存在差别;HCC组织中ALC1的阳性表达与EZH2表达正相关。
Objective To investigate the localization and correlation of chromatin helicase ALC1 and transcription factor EZH2 in hepatocellular carcinoma (HCC). Methods In this study, RT-PCR, immunofluorescence double labeling and immunohistochemistry were used to study the co-expression of ALC1 and EZH2 in hepatocellular carcinoma and hepatocellular carcinoma at mRNA and protein levels. Results The positive expression rates of ALC1 and EZH2 mRNA in hepatocellular carcinoma cells were detected. Immunofluorescence showed that the positive expression products of ALC1 and EZH2 localized in the nucleus. The positive rates of ALC1 and EZH2 in HepG2 cells were 78.0% and 89.2% The positive expression rate of EZH2 in HepG2 cells was 53.6% and 74.6%, respectively. The co-expression rate of EZH2 in HepG2 cells was 71.2% and that in Huh7 cells was 47.3%. The results of immunohistochemistry in hepatocellular carcinoma The positive expression of ALC1 was positively correlated with the positive expression of EZH2 (P <0.05), and its co-expression rate was 48.1%. Conclusions The positive rate and co-expression rate of ALC1 and EZH2 in each hepatocellular carcinoma cell line are different. The positive expression of ALC1 in HCC tissues is positively correlated with the expression of EZH2.
