目的:探讨人结肠癌及远癌组织中RUNX3 mRNA和RUNX3蛋白表达的临床病理意义.方法:用逆转录聚合酶链反应(RT-PCR)分别检测50例手术切除的结肠癌组织及远癌组织中RUNX3 mRNA的表达;用免疫组化方法(SP法)检测100例结肠癌及远癌组织中RUNX3蛋白的表达.结果:RUNX3 mRNA和RUNX3蛋白在远癌组织中的表达明显高于结肠组织[(0.28±0.05)vs(0.41±0.05),P<0.01;58%vs100%,P<0.01].RUNX3蛋白的表达与癌细胞分化程度,淋巴结转移以及临床分期(Dukes分期)明显相关(U=144,P<0.01;χ2=15.11,P<0.01;χ2=11.02,P<0.05)与患者性别、年龄及肿瘤部位无关.结论:RNX3基因的表达异常与结肠癌的发生、发展密切相关.
Objective: To investigate the clinicopathological significance of RUNX3 mRNA and RUNX3 protein expression in human colon cancer and distant cancer.Methods: Reverse transcription polymerase chain reaction (RT-PCR) was used to detect 50 cases of resected colon cancer tissues and distant cancer tissues RUNX3 mRNA was detected by immunohistochemistry (SP method) in 100 cases of colon cancer and distant cancer tissues.Results: The expression of RUNX3 mRNA and RUNX3 protein in distant cancer tissues was significantly higher than that in colon tissues [ (0.28 ± 0.05 vs 0.41 ± 0.05, P <0.01, 58% vs100%, P <0.01) .RUNX3 protein expression was significantly correlated with the degree of cancer cell differentiation, lymph node metastasis and clinical stage (Dukes stage) (U = 144, P <0.01, χ2 = 15.11, P <0.01, χ2 = 11.02, P <0.05) .Conclusion: The abnormal expression of RNX3 is closely related to the occurrence and development of colon cancer.
