以溶菌酶(LYS)作为模式蛋白,对聚乙二醇化蛋白质在不同阳离子交换填料中色谱行为进行了比较研究。考察了不同分子质量的聚乙二醇化溶菌酶(PEG-LYS)在4种阳离子树脂中的动态载样量的差异,同时比较了不同上样量对纯度和洗脱盐浓度的影响。研究结果显示PEG修饰能降低蛋白质和离子交换树脂的结合力,且随着PEG的分子质量的增加,PEG-LYS动态载样量降低。对相同分子质量的PEG-LYS,产物纯度以及PEG-LYS的洗脱盐浓度随着上样量的增加而降低,研究结果显示,PEG分子质量和上样量对纯化工艺的研究有重要意义。
The chromatographic behavior of pegylated proteins in different cation exchange media was compared with lysozyme (LYS) as a model protein. The dynamic loading capacity of PEG-LYS with different molecular weight was investigated in four cationic resins. The effects of different loadings on the purity and elution salt concentration were also compared. The results showed that the PEG modification can reduce the binding capacity of protein and ion exchange resin, and with the increase of molecular weight of PEG, PEG-LYS dynamic loading capacity decreased. The PEG-LYS with the same molecular weight, the product purity and the elution salt concentration of PEG-LYS decreased with the increase of sample loading. The results showed that the molecular weight of PEG and sample loading were of great significance to the purification process.
