目的建立新疆维吾尔民间常用药阿里红中总三萜酸及去氢硫色多孔菌酸的测定方法。方法采用香草醛-冰醋酸、高氯酸比色法,以药材中指标成分去氢硫色多孔菌酸为对照品,检测波长为554 nm,测定总三萜酸的量。采用Agilent Eclipse XDB-C18色谱柱(250 mm×4.6 mm,5μm),以乙腈-0.4%磷酸水溶液(61∶39)为流动相,242 nm为检测波长,柱温25℃,体积流量1.0 mL/min,高效液相色谱法测定去氢硫色多孔菌酸的量。结果阿里红中总三萜酸在0.040 4~0.242 4 mg/mL呈良好的线性关系(r=0.998 9),加样回收率均值为98.56%。HPLC法测定去氢硫色多孔菌酸在0.022 3~0.133 7 mg/mL线性关系良好(r=0.999 7),加样回收率均值为100.68%。结论两种方法均操作简便,重现性好,结果可靠,适用于阿里红药材中三萜成分的质量控制。
OBJECTIVE To establish a method for the determination of total triterpene acid and polysulfonic acid in polyuronic acid of common red drug Uighur in Xinjiang Uygur Autonomous Region. Methods Vanillin - acetic acid and perchloric acid colorimetric method was used to determine the total amount of triterpene acid. The detection wavelength was 554 nm. An Agilent Eclipse XDB-C18 column (250 mm × 4.6 mm, 5 μm) was used with acetonitrile-0.4% phosphoric acid solution (61:39) as the mobile phase at 242 nm as the detection wavelength at a column temperature of 25 ° C with a volumetric flow rate of 1.0 mL / min, the determination of the amount of polysulfonic acid polyglutamic acid by high performance liquid chromatography. Results The total triterpene acid in Ali red showed a good linear relationship (r = 0.998 9) between 0.040 4 and 0.242 4 mg / mL. The average recoveries were 98.56%. The linear relationship was determined by HPLC with 0.026 3 ~ 0.133 7 mg / mL deuterated polysulphone (r = 0.999 7). The average recovery was 100.68%. Conclusion Both methods are simple, reproducible, and reliable. They are suitable for the quality control of triterpene constituents in Alixia red.
