目的:建立毛细管电泳法测定麻杏石甘汤中钙镁含量的方法,并测定不同配伍麻杏石甘汤中钙镁的溶出量。方法:采用L9(34)正交设计,用毛细管电泳法测定钙镁的含量。电泳条件:背景缓冲液为6.4 mmol.L-1咪唑溶液(冰乙酸调pH至3.5);检测波长214 nm;分离电压10 kV;重力进样(8 cm×7 s);柱温室温。结果:钙离子测定的线性范围0.009 68~9.68mg.L-1,相关系数0.998 1,检测限0.002 1 mg.L-1,定量限0.007 1 mg.L-1,平均回收率100.4%;镁离子测定的线性范围为0.01~10 mg.L-1,相关系数为0.995 9,检测限为0.002 8 mg.L-1,定量限0.008 9 mg.L-1,平均回收率96.4%。结论:本法快速简便、灵敏度高,可作为麻杏石甘汤中钙镁含量的测定方法。麻杏石甘汤中麻黄和杏仁对钙的溶出量影响显著,炙甘草对钙的溶出量影响不显著;麻黄对镁的溶出量影响显著,而杏仁和炙甘草对镁的溶出量影响不显著。
Objective: To establish a method for the determination of calcium and magnesium in Maxingshigan Decoction by capillary electrophoresis and to determine the dissolution of calcium and magnesium in different compatibility Maxingshigan Decoction. Methods: L9 (34) orthogonal design was used to determine the content of calcium and magnesium by capillary electrophoresis. Electrophoresis conditions: 6.4 mmol.L-1 imidazole solution (pH adjusted to 3.5 with glacial acetic acid) as the background buffer; detection wavelength 214 nm; separation voltage 10 kV; gravity injection (8 cm × 7 s); Results: The linear range of the determination of calcium was 0.009 68 ~ 9.68mg.L-1, the correlation coefficient was 0.998 1, the limit of detection was 0.002 1 mg.L-1, the limit of quantification was 0.007 1 mg.L-1, the average recovery was 100.4% The linear range was 0.01-10 mg.L-1, the correlation coefficient was 0.995 9, the limit of detection was 0.002 8 mg.L-1, the limit of quantification was 0.008 9 mg.L-1, and the average recovery was 96.4%. Conclusion: The method is rapid, simple and sensitive and can be used as a method for determination of calcium and magnesium in Maxingshigan Decoction. Maxing Shigan Decoction Ephedrine and almond have a significant effect on the dissolution of calcium, Baked Licorice on the dissolution of calcium has no significant effect; Ephedra significantly affected the dissolution of magnesium, almond and Zhigancao on the dissolution of magnesium was not significant .
