目的:建立UPLC-MS/MS测定大鼠尾静脉注射荭草花提取物后的大鼠血浆中3种成分(原儿茶酸、山柰素-葡萄糖苷、檞皮苷)含量的分析方法,并研究这3种成分在大鼠体内药代动力学特征。方法:采用Waters BEH C18(50 mm×2.1 mm,1.7μm)色谱柱;流动相为0.1%甲酸乙腈-0.1%甲酸溶液,梯度洗脱;质谱采用电喷雾电离源(ESI);扫描方式为多反应离子监测(MRM)。结果:静脉注射荭草花提取物后原儿茶酸、山柰素-葡萄糖苷、槲皮苷在大鼠体内的半衰期分别为(41.9±12.3)min、(71.3±56.8)min和(90.3±74.8)min,达峰浓度(1.7±0.7)μg/m L、(2.1±0.9)μg/m L和(8.7±3.7)μg/m L。结论:实验结果表明建立的方法快速、准确、灵敏,精密度回收率均符合生物样品的测定要求,适用于原儿茶酸、山柰素-葡萄糖苷、檞皮苷3个成分在大鼠血浆中浓度的测定,3个成分在大鼠体内的药动学特征均符合二室模型。
OBJECTIVE: To establish a method for the determination of three components in rat plasma (protocatechuic acid, kaempferol-glycoside, kaempferide) by UPLC-MS / MS in rat tail vein injection The pharmacokinetic profiles of these three components in rats were studied.
