目的:探讨威柏膏外敷治疗急性痛风性关节炎模型大鼠的药效及可能的作用机制。方法:雄性Wist-ar大鼠随机分为正常对照组(A)、模型组(B)、秋水仙碱组(C)、威柏膏外敷组(D)、秋水仙碱加威柏膏组(E)。观察大鼠足功能变化,检测其受试关节周径、滑膜病理改变,以及关节滑膜组织中白介素1-β(IL-1β)、5-羟色胺(5-HT)和TLR-4(Toll-like receptor 4)的相对表达量。结果:与模型组比较,各治疗组均可减少炎性细胞浸润,并能显著降低IL-1β、TLR-4的表达。E组较D组降低IL-1β的表达更为显著;E组能显著降低5-HT的相对表达含量。结论:威柏膏外敷能下调大鼠急性痛风性关节炎的炎性反应,止痛消炎,其机制可能是降低了滑膜组织中IL-1β、TLR-4及5-HT的表达;威柏膏联合秋水仙碱比单用两者之一的效果更好。
Objective: To investigate the efficacy and possible mechanism of victoir cream in the treatment of acute gouty arthritis in rats. Methods: Male Wist-ar rats were randomly divided into normal control group (A), model group (B), colchicine group (C), Weibai cream external application group (D), colchicine plus weibai cream group E). The changes of foot function in rats were observed. The changes of joint circumference, synovial pathological changes, interleukin-1β (IL-1β), 5-HT and TLR-4 -like receptor 4). Results: Compared with the model group, each treatment group could reduce inflammatory cell infiltration and significantly reduce the expression of IL-1β, TLR-4. E group compared with D group decreased IL-1β expression more significant; E group can significantly reduce the relative expression of 5-HT content. Conclusion: Weibai Ointment can down-regulate the inflammatory reaction and relieve pain of acute gouty arthritis in rats. Its mechanism may be to decrease the expression of IL-1β, TLR-4 and 5-HT in synovial tissue. Colchicine is more effective than either alone.
