以梨砧木OH×F51为试材,对其茎尖组织培养和叶片愈伤组织诱导进行了研究,以获得OH×F51离体培养的最佳途径。结果表明:适合OH×F51茎尖离体培养的培养基是MS+6-BA 2.0mg/L+NAA 0.2mg/L+蔗糖30g/L+琼脂6g/L,有较高的成苗率;OH×F51叶片愈伤组织诱导的最佳培养基是NN69+TDZ 3.0mg/L+IBA 1.0mg/L+蔗糖30g/L+琼脂6g/L,叶片放置方式对愈伤组织诱导有一定影响,叶片的远轴面接触培养基,可以获得较高的出愈率。
Taking rootstock OH × F51 as experimental material, the shoot tip culture and leaf callus induction were studied in order to obtain the best way of OH × F51 culture in vitro. The results showed that the medium for culturing OH × F51 in vitro was MS + 6-BA 2.0 mg / L + NAA 0.2 mg / L + sucrose 30 g / L + agar 6 g / L, The optimal culture medium for induction of callus from leaves of F51 was NN69 + TDZ 3.0 mg / L + IBA 1.0 mg / L + sucrose 30 g / L + agar 6 g / L. The leaf placement mode had a certain influence on the callus induction. Surface contact with the medium, you can get a higher cure rate.
