目的探讨抑癌基因在乳腺细胞系的表达及遗传学调控.方法以11种乳腺癌细胞系为材料,乳腺非致瘤细胞株(MCF-10A)作为参照,分别于 RNA 水平及蛋白质水平分析 MARVELD1内源性表达,并筛选出明显低于 MCF-10A 的乳腺细胞系.经去甲基化诱导剂与组蛋白去乙酰化酶抑制剂处理后,检测MARVELD1的表达水平变化.结果 MARVELD1基因于12种乳腺细胞系内源性表达,其中 SK-BR-3中的 MARVELD1表达明显低于非致瘤乳腺上皮细胞 MCF10A;去甲基化诱导剂5-Aza-CdR 处理 SK-BR-3后,MARVELD1基因表达明显升高.结论 MARVELD1的基因表达可能受启动子甲基化与组蛋白乙酰化的调控.“,”Objective: Learn the express and genetic regulation of anti-oncogene in breast cel lines. Methods: There were 11 cases of breast cel line as material and breast non-tumorigenic cel lines (MCF-10A) as reference. Analyze the endogenous expression of MARVELD1 in RNA and protein level, and choose the breast cel lines lower than MCF-10A. Check the expression of MARVELD1 after being treated by demethylation induced agent and histone deacetylase inhibitors. Results: In the endogenous expression of MARVELD1 gene in 12 cases of breast cel lines, the expression of MARVELD1 from SK-BR-3 was obviously lower than nononcogenic mammary epithelial cels MCF10A. The expression of MARVELD1 was obviously higher after being treated by demethylation induced agent 5-Aza-CdR. Conclusion: The expression of MARVELD1 may be regulated by promoter methylation and histone acetylation.
