目的:建立离体牛蛙肺泡膜模型并初步探讨胰岛素的肺吸收机制。方法:建立离体牛蛙肺泡膜模型,对模型的完整性、通透性及影响因素等进行考察,并初步研究胰岛素的肺吸收机制。结果:在4 h内,膜的紧密性和通透性良好,在一定范围内胰岛素透过牛蛙肺泡膜是非浓度、非方向依赖性的,且单体较六聚体透过率高。结论:离体牛蛙肺泡膜模型可作为评价胰岛素肺吸收的体外模型。经比较,单体胰岛素可能比六聚体胰岛素肺吸收好。
OBJECTIVE: To establish a model of isolated bullfrog alveolar membrane and to investigate the mechanism of insulin absorption. Methods: The model of alveolar membrane in vitro was established and the integrity, permeability and influencing factors of the model were investigated. The lung absorption mechanism of insulin was also studied. Results: Within 4 h, the tightness and permeability of the membrane were good. In a certain range, the transmission of insulin through the bullfrog alveolar membrane was non-concentration and non-directional, and the monomer was more permeable than the hexamer. CONCLUSION: The isolated bullfrog alveolar membrane model can be used as an in vitro model to evaluate the lung absorption of insulin. By comparison, monomeric insulin may absorb better than the hexameric insulin lung.
