抗哇巴因多克隆抗体的制备及免疫周期和剂量的研究

制备抗哇巴因多克隆抗体 ,探讨最佳的免疫周期及免疫剂量。应用哇巴因 (ouabain) -牛血清白蛋白 (BSA)结合物为抗原免疫家兔 ,每周从兔耳缘静脉采血 ,监测哇巴因特异抗体的产生 ;以0 .2 5mg/次、1mg/次、1.75mg/次、2 mg/次四种不同免疫剂量进行最佳免疫剂量的研究。 5个月后颈动脉取血 ,进行抗体效价、亲和力及特异性的检测。结果显示免疫间隔为 6周、免疫剂量为 1.75mg/次时可以获得高效价、高亲和力和高特异性的哇巴因抗血清。

Preparation of anti-ouabain polyclonal antibody, to explore the best immune cycle and immune dose. Rabbit was immunized with ouabain-bovine serum albumin (BSA) conjugate and blood was collected weekly from rabbits’leaf vein to monitor ouabain-specific antibody production. At 0.52 mg / / Times, 1.75mg / time, 2mg / times the four different doses of immune best dose for the study. Carotid blood was taken 5 months later for antibody titer, affinity and specificity testing. The results showed that the immunization interval was 6 weeks and the immunization dose was 1.75mg / time, the high potency, high affinity and high specificity of ouabain antiserum could be obtained.
